چکیده

12 13 The authors declare that no competing interests exist. 14 2 Abstract 15 Autophagy is a major pathway for the clearance of harmful material from the cytoplasm. 16 During autophagy cytoplasmic material is delivered into the lysosomal system by organelles 17 called autophagosomes. Autophagosomes form in a de novo manner and, in the course of their 18 formation, isolate cargo material from the rest of the cytoplasm. Cargo specificity is conferred 19 by autophagic cargo receptors that selectively link the cargo to the autophagosomal 20 membrane decorated with ATG8 family proteins such as LC3B. Here we show that the human 21 cargo receptor p62/SQSTM-1 employs oligomerization to stabilize its interaction with LC3B 22 and linear ubiquitin when they are clustered on surfaces. Thus, oligomerization enables p62 23 to simultaneously select for the isolation membrane and the ubiquitinated cargo. We further 24 show in a fully reconstituted system that the interaction of p62 with ubiquitin and LC3B is 25 sufficient to bend the membrane around the cargo.

The higher-order molecular organization of p62/SQSTM1

The multifunctional signaling adaptor and selective autophagy receptor p62/SQSTM1 is commonly found in dense light-microscopic loci of eukaryotic cells. Recently, Ciuffa et al. demonstrated that p62 is able to form organized polymers of helical symmetry once purified and reconstituted in the test tube [1]. In selective autophagy, p62 acts both as a substrate and as a receptor to bridge LC3 atta...

MAP1B Interaction with the FW Domain of the Autophagic Receptor Nbr1 Facilitates Its Association to the Microtubule Network

Selective autophagy is a process whereby specific targeted cargo proteins, aggregates, or organelles are sequestered into double-membrane-bound phagophores before fusion with the lysosome for protein degradation. It has been demonstrated that the microtubule network is important for the formation and movement of autophagosomes. Nbr1 is a selective cargo receptor that through its interaction wit...

PpAtg30 tags peroxisomes for turnover by selective autophagy.

Autophagy, an intrinsically nonselective process, can also target selective cargo for degradation. The mechanism of selective peroxisome turnover by autophagy-related processes (pexophagy), termed micropexophagy and macropexophagy, is unknown. We show how a Pichia pastoris protein, PpAtg30, mediates peroxisome selection during pexophagy. It is necessary for pexophagy, but not for other selectiv...

Mechanism of cargo-directed Atg8 conjugation during selective autophagy

Selective autophagy is mediated by cargo receptors that link the cargo to the isolation membrane via interactions with Atg8 proteins. Atg8 proteins are localized to the membrane in an ubiquitin-like conjugation reaction, but how this conjugation is coupled to the presence of the cargo is unclear. Here we show that the S. cerevisiae Atg19, Atg34 and the human p62, Optineurin and NDP52 cargo rece...
